When it comes to filming, actors are required to follow the script, unless they discuss with the Director ahead of time. Of course, when an actor or actress are auditioning for a film, they should already have learned about it ahead of time, so they know what they are going into. 

But, when it comes to Romance scenes in films or TV shows, how exactly is it done? Many directors do things differently, which depends on their boundaries, the actors boundaries (Which sometimes, aren't set), and what they are comfortable with.

There are times where an actor or actress are not comfortable, so they find a double, and avoid showing the face as to make it less noticeable that they aren't the same person.

But, when it does come to the original actor and actress having to deal with a romance scene, there is a lot that goes into it. From the chemistry, to the editing (And sometimes, the kissing sounds are re-made by the sound editor and Foley Artists), to the idea of having to do whatever they are doing, in front of a crew that can span from 4-60 people. At times with more involved scenes, the scenes are done on a closed set with limited number of people. (Not to mention the audience who will end up watching the film). Also, with filming, the actors/actresses follow scripts/verbal directions and do what looks good to the camera and not necessarily what may be comfortable to them. 

It also depends on the relationship they have with their co-worker with whom they are doing the scenes with. From not getting along, or they do get along, but see their relationship as brother/sister. Some actors and actresses tend to say that they do, in fact, enjoy doing romantic scenes, but there are those who say they don't. 

For example: Emma Watson and Rupert Grint in Harry Potter. They had a very strong sister/brother relationship, (According to interviews) and when it came to the last movie where they kiss, they had many bloopers and were very nervous and slightly uncomfortable, due to the non-romantic chemistry they had while on set.
